Yemen: thousands demand retrieval of stolen assets

SANAA, Yemen (AP) ? Thousands have taken to the streets of the Yemeni capital, Sanaa, demanding the recovery of assets they say were stolen under the country's former authoritarian ruler, Ali Abdullah Saleh.

The protesters marched to demand return of millions of dollars they allege Saleh and his family stole during his years in power. Saleh, known as a wealthy ruler who led one of the Arab world's poorest countries, stepped down after a yearlong popular uprising which forced him to relinquish power in return for immunity from prosecution.

The Friday protests come a week after several nations backing Yemen's political transition pushed for sanctions against Saleh loyalists and other groups for undermining the country's shift to democracy. The non-military sanctions include freezing financial assets or travel ban.

Rowling's first novel for adults goes on sale

LONDON (AP) ? British bookshops opened their doors early Thursday and some grown-up Harry Potter fans lined up overnight as J.K. Rowling launched her long-anticipated first book for adults, "The Casual Vacancy."

The lines were shorter and the wizard costumes missing, but the book was published to some of the same fanfare that greeted each Potter tome, with stores wheeling out crates of the books precisely at 8 a.m.

Published five years after the release of the last book in the boy wizard saga, "The Casual Vacancy" is already at No. 1 on Amazon's U.S. chart, and bookmaker William Hill put 2/1 odds on it outselling "Harry Potter and the Deathly Hallows," which shifted 2.6 million copies in Britain on its first day.

Many of the early buyers were Harry Potter fans who, like the author, have moved on to more adult fare.

"I just like how much excitement there is about a book," said 23-year-old Grace Proctor, a "massive" Potter fan who was first to buy the book at one London store.

"I think there are going to be people who will hate it just because they can hate it," she said. "But she's such an amazing writer, I don't think she can go wrong."

A story of ambition and rivalry set in the fictional English village of Pagford, the novel recounts the civic warfare sparked when the unexpected death of a town official leaves a vacancy on the governing body.

Rowling has said she was aiming for Dickensian sweep in the multi-character saga, whose doses of sex, satire and swearing mark a distinct departure from wholesome Harry.

Reviews have been mixed. The Associated Press judged it a challenging but rewarding read full of emotion and heart, while The Guardian newspaper said it was "no masterpiece, but it's not bad at all: intelligent, workmanlike, and often funny." Several reviewers felt the lack of easily likable characters might be off-putting to readers.

It's likely nothing Rowling publishes will ever match the success of the Potter books, which have sold more than 450 million copies around the world.

But booksellers are confident "The Casual Vacancy" will be one of the year's best-sellers, whatever reviewers say.

"A lot of children have grown up with Harry Potter. They're now adults who love books," said Susan Sinclair, divisional manager for the Foyles bookstore chain.

"I think it's going to be a really big seller at Christmas. It'll be an easy gift ? but also a good one."

Edmonds man realizes dream of starting online radio station

In June 2011, Edmonds resident Michael Raley walked away from a successful career in radio advertising, with one mission in mind: to build a radio station of his own.

But Raley wasn?t looking to start just any station. He was tired of commercial radio?s lack of creativity ? both in terms of the music it played and the advertising it ran. He wanted a station that supported and celebrated local music, musicians and businesses. And he wanted to make the broadcasts available on a digital platform.

This week, Raley?s vision become a reality with the official launch of Jet City Stream. The station?s focus is on local music, which includes finding and featuring local bands.? Since April, when the station first went ?on the air? from its headquarters in Seattle?s Georgetown neighborhood, Jet City staff have been working to build their brand at various community music events, participating in the Capitol Hill Block Party, Bumbershoot and the Crooked Chinook Music Festival, among others. Jet City has also hosted several smaller events at its studio location inside the former Rainier Brewery-turned-Tully?s-Coffee headquarters on Airport Way South.

The most important room in the Jet City studio suite is probably the server room, complete with a 10.5 mm fiber optic cable ? ?the biggest you can get,? Raley said. ?We wanted to push the audio because EQ (equalization) on the Internet is average at best. So what we did was we created two streams for one fully-equalized path. When you listen to it online, it balances out highs and lows. You can hear total separation in what we do. ?

In addition, the station has a functional air studio where they have held Jet City Stream Sessions with local bands including Reignwolf, Walking Papers, Father John Misty and Fly Moon Royalty.

The station also plans to feature famous names as they come through the Puget Sound region for a concert. ?Our goal is to get those bands to come down to the station, do an acoustic or electric set with us and leave us with those gems ? and then we can play those songs for eternity because they were recorded with us,? Raley said.

He envisions eventually expanding the concept to 25 cities nationwide, creating an online network of Internet stations, each with the same focus on local music and community events.

Part of what drives the 50-year-old Raley is his memories of just how good radio used to be during his childhood and teen years, when a station would play a diverse mix? of music ? everything from Glen Campbell and Elvis Presley to Roberta Flack and Queen.? Today, Raley noted, ?there are five major (music) labels and they control 50 major bands, so they fund those bands and that?s it.? As a result, many great bands and their music are left in the collective dust.

?We just wanted to take the great things radio did and combine it with today?s technology of music discovery,? he said.

Most people in the Internet radio business are ?either doing it in their basement in some bad audio way or they are doing Pandora style, which is not radio ? it?s just an iPod online where you can click a song,? Raley said. ?It?s not radio DJs. It doesn?t have localism.?

Raley himself is a former Edmonds restaurant manager ? he worked at Anthony?s Home Port from 1985-1995? who has been in love with music since he started playing guitar in a rock band at age 16.

After encouragement from an acquaintance, he moved into radio sales at age 37, starting with KISS-FM and eventually ending up in Los Angeles before moving back to Edmonds and a sales job with Seattle-based Clear Channel.

However, he grew tired of the type of advertising that was being aired and added that he ?felt that the best music in the world wasn?t getting played on the radio,? due to what he called ?the corporate rule of record labels.?

As he began building his new station, Raley?s top two priorities were his staff? ? ?finding the best people in Seattle? ?? and also securing what he described as ?an iconic location: a place where a band could bring their record down and see the DJs and meet them.?

His success so far is reflected in both the choice of the station?s home ? the hip yet comfortable former Rainier Brewery office suite? ? and his equally impressive hires: Shawn Stewart of 103.7 The Mountain fame is Jet City Stream?s content director and is on the air fron 10 a.m.-3 p.m. Marco Collins, who broadcasts from 3-7 p.m., is a member of the Rock and Roll Hall of Fame and best known as the DJ and Music Director at Seattle?s 107.7? ?The End? during the 1990?s grunge era. Running the station?s marketing is Joe Hammill, former ?The End? promotions director.? And Jet City?s vice president of programming is Becky Brenner, who was program director at KMPS for 16 years.

The well-known names continue as part of this week?s official Jet City Stream launch. On Friday, Collins will be joined by Seattle radio legend Pat O?Day, who will co-host the Top 20 Seattle Songs, starting at 3 p.m. According to Raley, the combination was a natural fit: The two personalities have made a name for themselves by discovering the best sounds from Seattle from the ?60s (O?Day, at KJR 950) and the ?90s (Collins at 107.7).?and both men are in the Rock and Roll Hall of Fame.

The Seattle Top 20 Countdown is a weekly feature hosted by Collins that covers the best-selling albums from Seattle artists, based on local record store sales.

MOGADISHU, Somalia --?Troops launched an amphibious assault before dawn Friday on the al-Qaida militant group al-Shabab's last stronghold in Somalia.

Other African Union forces were traveling overland to link up with the joint Kenyan-Somali force in the port city of Kismayo.

The commander of the U.N-backed African Union troops, Lt. Gen. Andrew Gutti, said the aim was to "liberate the people of Kismayo to enable them to lead their lives in peace, stability and security."

Col. Cyrus Oguna, the Kenyan military's top spokesman, said the attack met minimal resistance, but al-Shabab denied that the city had fallen and said fighting was taking place.

Oguna told The Associated Press that al-Shabab, which formally merged with al-Qaida in February, had incurred "heavy losses" but that Kenyan forces have not yet had any injuries or deaths.

"We came from the beach side and we're moving towards the main city. Our surveillance aircraft are monitoring every event taking place on the ground," he told Reuters.

"For now, we're not everywhere. We've taken a large part of it without resistance, I don't see anything major happening," he said.

Residents in Kismayo, a city of about 193,000 people, contacted by The Associated Press said that Kenyan troops had taken control of the port, but not the whole city.

"Al-Shabab fighters are on the streets and heading toward the front line in speeding cars. Their radio is still on the air and reporting the war," resident Mohamed Haji told The Associated Press. Haji said that helicopters were hitting targets in the town in southeastern Somalia.

At an international one-day summit Britain's Prime Minister David Cameron said the world would "pay a price" if it fails to help Somalia overcome terrorism, piracy and starvation. ITV's Lee Comley reports.?

Another resident, Ismail Suglow, told Reuters that he could hear shelling from the ships and that the rebels were responding with anti-aircraft guns.

"We saw seven ships early in the morning and now their firing looks like lightning and thunder. Al-Shabab have gone towards the beach. The ships poured many AU troops on the beach," he added.

Expert: War on terror at 'critical' point as al-Qaida looks to regroup in Africa

On Thursday, residents said planes had dropped leaflets on Kismayo warning civilians to evacuate within 24 hours, Reuters reported.?More than 10,000 residents fled Kismayo in the last several weeks.

Resident Faduma Abdulle said Friday that she is now leaving too.

She said al-Shabab made an announcement on its radio station Friday to trick residents into moving toward the invading troops.

"They told residents through their radio to loot a Kenyan ship that washed up on the coast, but instead the residents who rushed there were attacked by helicopters," she said. "Some of them have died but I don't know how many. The situation is tense and many are fleeing. It's a dangerous situation."

A U.S. military spokesman, Lt. Cdr. Dave Hecht, said the U.S. Africa Command, known as AFRICOM, is closely monitoring the situation but that "we are not participating in Kenya's military activities in the region."

Militants: Taking city not 'a piece of cake'
Al-Shabab said it would not give up Kismayu easily.

"Going into Kismayo is not a piece of cake. We are still fighting them on the beach where they landed," Sheik Abdiasis Abu Musab, al-Shabab's spokesman for military operations, told Reuters on Friday. "For us, this is just the beginning, our troops are spread everywhere."

Oguna said the assault is part of a four-prong attack involving Kenyan forces currently in villages outside Kismayo.

The amphibious assault landed between 10:30 p.m. Thursday and 2 a.m. Friday local time (3:30 p.m. and 7:30 p.m. Thursday ET)?, he said. Some of the troops had night-vision goggles, he said.

African Union troops pushed al-Shabab out of Mogadishu in August 2011, ending four years of control of the capital by the fighters.

The Ugandan and Burundian troops that make up the bulk of the African Union force in Mogadishu have slowly been taking control of towns outside of Mogadishu.

The expanding control by AU troops sent al-Shabab fighters fleeing south toward Kismayo, north to other regions of Somalia and across the Gulf of Aden to Yemen, according to American and African Union officials.

Kenya police: Imminent attack by suicide bombers thwarted

Al-Shabab still holds sway across many small, poor villages of southern Somalia. The loss of Kismayo would be significant.

The militants taxed goods coming into its port. Al-Shabab lost its major source of financing last year when it was pushed out of Bakara market in Mogadishu, where it also charged taxes.

The march toward Kismayo by the Kenyan forces has been nearly a year in the making.

Masked 'goons' kill at least 17 in attacks on churches in Kenya

Kenyan troops entered Somalia last October after a string of kidnappings inside neighboring Kenya, including of Westerners in and around the beach resort town of Lamu, which is also seeing the construction of a new port and could one day be final point of a new oil pipeline from South Sudan.

Kenyan forces were bogged down by rain and poor roads for months but have making slow and steady progress toward Kismayo in recent weeks.

Slave rebellion is widespread in ants

ScienceDaily (Sep. 26, 2012) ? Enslaved worker ants kill the offspring of their parasites and thereby improve the chances of survival for their neighboring relatives.

Ants that are held as slaves in nests of other ant species damage their oppressors through acts of sabotage. Ant researcher Professor Dr. Susanne Foitzik of Johannes Gutenberg University Mainz (JGU) in Germany first observed this "slave rebellion" phenomenon in 2009. According to the latest findings, however, this behavior now appears to be a widespread characteristic that is not limited to isolated occurrences. In fact, in three different populations in the U.S. states of West Virginia, New York, and Ohio, enslaved Temnothorax longispinosus workers have been observed to neglect and kill the offspring of their Protomognathus americanus slavemakers rather than care for them. As a result, an average of only 45 percent of the parasite's offspring survived. This presumably reduces the strength of the parasites in the area and thereby increases the chances of survival for the neighboring colonies populated by the slave ants' relatives.

More than half of all animal species live in parasitic relationships, i.e. they exploit their so-called hosts. From the perspective of evolutionary history, the American slave-making ant Protomognathus americanus is an old social parasite that is entirely dependent on other ant species for its survival. Slave workers have to care for the brood in parasite nests, bring food to their masters and feed them, and even defend the nest.

These ants become slaves when workers from the slave-making ant colony attack the nests of the host species Temnothorax longispinosus, kill the adult ants, and steal the brood. Back in the masters' nest, which can be located in hollow acorns, nutshells, or twigs, the brood care behavior of the emerging slave workers is exploited to the advantage of the slavemaker species. As Susanne Foitzik and her work group have shown, the enslaved worker ants feed and clean the larvae, thereby raising the offspring of their social parasite -- but only up to a certain point.

"Probably at first the slaves cannot tell that the larvae belong to another species," explains Foitzik. As a result, 95 percent of the brood survives the larval stage. But the situation changes as soon as the larvae pupate. "The pupae, which already look like ants, bear chemical cues on their cuticles that can apparently be detected. We have been able to show that a high fraction of the slavemaker pupae are killed by slave workers." The pupae are either neglected or actively killed by being attacked and torn apart. Several slaves at once may assault a pupa, which is unable to move or defend itself during the pupal stage and is also not protected by a cocoon.

In parasite nests in West Virginia, only 27 percent of the pupae survived, and in the New York colonies, only 49 percent. In Ohio, the survival chances of the American slave-making ant was a bit higher at 58 percent, but this figure is still well below the survival rate of 85 percent for host pupae in their own free-living nests. "The enslaved workers do not directly benefit from the killings because they do not reproduce," explains Susanne Foitzik. But, through the killing of slavemaker offspring, their neighboring relatives -- which might very well be the sisters of the worker slaves -- indirectly benefit as their chances of survival are increased. Slavemaker colonies damaged by slave rebellions grow slower and smaller slave-making colonies conduct fewer and less destructive slave raids.

The large differences in the death rates in colonies from different regions fits to predictions derived from the geographic mosaic theory of co-evolution. This theory claims that populations differ because they are subjected to different local selection pressures and because they possess different attack or defense traits originated through mutations, which in turn means that evolution can go in different directions in different geographic areas. While the host ants in New York are very aggressive and often successfully thwart slave raid attempts, the hosts in West Virginia profit more from the slave rebellion behavior because, as genetic analyses have shown, the neighboring colonies are more often close relatives to the rebelling slaves.

This study on the evolution of slave rebellion has been financed since October 2011 by the project "The evolution of resistance and virulence in structured populations" funded by the German Research Foundation.

Next month, Valve is inviting 1,000 fortunate gamers to take part in the first external beta test of Steam for Linux. The trial includes the service itself and one game (which we're taking to be Left 4 Dead 2) that'll run on systems running Ubuntu 12.04 and above. Users won't get their hands on any other Valve titles, or Big Picture Mode, and the company is asking that only experienced Linux users get in touch -- novices are politely asked to wait for a subsequent release. It'll offer up details of how to sign up shortly, and given the love for the Valve's other products, we suspect it'll be a little oversubscribed.

Making it easier to make stem cells: Kinase inhibitors lower barrier to producing stem cells in lab

ScienceDaily (Sep. 25, 2012) ? The process researchers use to generate induced pluripotent stem cells (iPSCs) -- a special type of stem cell that can be made in the lab from any type of adult cell -- is time consuming and inefficient. To speed things up, researchers at Sanford-Burnham Medical Research Institute (Sanford-Burnham) turned to kinase inhibitors. These chemical compounds block the activity of kinases, enzymes responsible for many aspects of cellular communication, survival, and growth.

As they outline in a paper published September 25 in Nature Communications, the team found several kinase inhibitors that, when added to starter cells, help generate many more iPSCs than the standard method. This new capability will likely speed up research in many fields, better enabling scientists around the world to study human disease and develop new treatments.

"Generating iPSCs depends on the regulation of communication networks within cells," explained Tariq Rana, Ph.D., program director in Sanford-Burnham's Sanford Children's Health Research Center and senior author of the study. "So, when you start manipulating which genes are turned on or off in cells to create pluripotent stem cells, you are probably activating a large number of kinases. Since many of these active kinases are likely inhibiting the conversion to iPSCs, it made sense to us that adding inhibitors might lower the barrier."

According to Tony Hunter, Ph.D., professor in the Molecular and Cell Biology Laboratory at the Salk Institute for Biological Studies and director of the Salk Institute Cancer Center, "The identification of small molecules that improve the efficiency of generating iPSCs is an important step forward in being able to use these cells therapeutically. Tariq Rana's exciting new work has uncovered a class of protein kinase inhibitors that override the normal barriers to efficient iPSC formation, and these inhibitors should prove useful in generating iPSCs from new sources for experimental and ultimately therapeutic purposes." Hunter, a kinase expert, was not involved in this study.

The promise of iPSCs

At the moment, the only treatment option available to many heart failure patients is a heart transplant. Looking for a better alternative, many researchers are coaxing stem cells into new heart muscle. In Alzheimer's disease, researchers are also interested in stem cells, using them to reproduce a person's own malfunctioning brain cells in a dish, where they can be used to test therapeutic drugs. But where do these stem cells come from? Since the advent of iPSC technology, the answer in many cases is the lab. Like their embryonic cousins, iPSCs can be used to generate just about any cell type -- heart, brain, or muscle, to name a few -- that can be used to test new therapies or potentially to replace diseased or damaged tissue.

It sounds simple enough: you start with any type of differentiated cell, such as skin cells, add four molecules that reprogram the cells' genomes, and then try to catch those that successfully revert to unspecialized iPSCs. But the process takes a long time and isn't very efficient -- you can start with thousands of skin cells and end up with just a few iPSCs.

Inhibiting kinases to make more iPSCs

Zhonghan Li, a graduate student in Rana's laboratory, took on the task of finding kinase inhibitors that might speed up the iPSC-generating process. Scientists in the Conrad Prebys Center for Chemical Genomics, Sanford-Burnham's drug discovery facility, provided Li with a collection of more than 240 chemical compounds that inhibit kinases. Li painstakingly added them one-by-one to his cells and waited to see what happened. Several kinase inhibitors produced many more iPSCs than the untreated cells -- in some cases too many iPSCs for the tiny dish housing them. The most potent inhibitors targeted three kinases in particular: AurkA, P38, and IP3K.

Working with the staff in Sanford-Burnham's genomics, bioinformatics, animal modeling, and histology core facilities -- valuable resources and expertise available to all Sanford-Burnham scientists and the scientific community at large -- Rana and Li further confirmed the specificity of their findings and even nailed down the mechanism behind one inhibitor's beneficial actions.

"We found that manipulating the activity of these kinases can substantially increase cellular reprogramming efficiency," Rana said. "But what's more, we've also provided new insights into the molecular mechanism of reprogramming and revealed new functions for these kinases. We hope these findings will encourage further efforts to screen for small molecules that might prove useful in iPSC-based therapies."
